GMP Vial Filler (or GMP-compliant Vial Filling Machine) is a specialized piece of pharmaceutical equipment designed for the aseptic filling of sterile vials with liquid or lyophilized (freeze-dried) drug products, such as injectables, vaccines, or biologics. It adheres to Good Manufacturing Practice (GMP) standards, which are regulatory guidelines enforced by bodies like the FDA, EMA, and others to ensure product quality, safety, and sterility by minimizing contamination risks during production. These machines are essential in pharmaceutical, biotechnology, and contract manufacturing organizations (CMOs) for handling small to large batches, from R&D/clinical trials to commercial-scale operations.
Built with cGMP compliance in mind, this machine ensures minimal human intervention, reduced contamination risk, and consistent product quality. It typically features a PLC-based control system with touchscreen HMI, advanced dosing technologies (peristaltic pumps, servo piston pumps, or powder dosing systems), and robust construction suitable for ISO Class 5 / Grade A cleanroom environments, including compatibility with RABS or isolators.

The incoming dry vials (sterilized and siliconized) are fed through the unscrambler and suitably guided on the moving Delrin slat conveyor belt at the required speed of the correct placement below filling unit.
The filling unit consists of Filling Head, Syringes & Nozzles; Syringes & Nozzles are made of AISI SS 316L materials. A Star Wheel is provided which holds the vial during filling operation. A sensor is provided for ”No Vial-No Filling” operation. Syringes are mounted on eccentric block and driving through bottom main gear box. Volume can be increase & decrease by increasing or decreasing stroke length of piston as per filling size. Syringes is having non return valve for sucking and delivering situation to avoid volume variation. Liquid reaches to filling nozzles through silicon transparent pipe. Nozzles having up & down movement with help of cam mechanism, and it come down when vial hold by starwheel for filling operation and starts filling when it starts to move up and after completion of filling operation, starwheel delivers two vials together on conveyor for rubber stoppering operation.
Stoppering unit consist of Star wheel, Vibrator unit and Bowl. The sterilized and siliconized rubber stoppers stored in the vibrator bowl moves vertically to the rubber stopper chute. The vial is hold firmly by starwheel, which will be in continuous rotary motion and vial which is coming from filling unit get in to starwheel and move in rotary direction along with starwheel, during movement it picks up the rubber stopper from the exit end of the chute and rubber stopper pressed by fix single pressing roller, which is spring loaded. Then vial further move on conveyor and collected on out feed starwheel for next sealing operation.

GMP vial fillers are typically integrated into cleanroom environments (e.g., ISO Class 5/Class A laminar flow) and often housed in isolators or Restricted Access Barrier Systems (RABS) for enhanced sterility. Here’s a breakdown of their core aspects:
A GMP Vial Filler is a fully automatic aseptic vial filling machine designed and built according to cGMP (Current Good Manufacturing Practice) standards. It is used for precise filling, stoppering, and sealing of sterile liquids or powders into injection vials. Equipped with PLC control and HMI, this machine ensures high accuracy, minimal contamination risk, and full compliance with FDA, EU Annex 1, and WHO regulatory requirements.
The key advantages include:
It is ideal for manufacturing high-quality injectable products safely and efficiently.
A GMP Vial Filler is constructed with AISI SS 316L product contact parts, crevice-free design, and mirror-polished surfaces for easy sterilization. It operates in ISO Class 5 / Grade A environments and supports Restricted Access Barrier Systems (RABS) or isolators. Features such as “No Vial – No Fill” interlock, automatic rejection, and validated cleaning procedures help maintain sterility while meeting strict regulatory documentation and validation requirements.
Production capacity of a GMP Vial Filler typically ranges from 60 to 300+ vials per minute, depending on the number of filling heads, vial size (2 ml to 100 ml), and whether it is processing liquid or powder formulations. The PLC-controlled system allows precise speed adjustment while maintaining consistent accuracy and GMP compliance across different batch sizes.
A GMP Vial Filler is specifically engineered for pharmaceutical companies that require strict regulatory compliance, audit readiness, and long-term reliability. Unlike standard machines, it offers validated design, superior material quality, complete documentation support, better data integrity, and lower risk of regulatory observations or product recalls. It is the preferred choice for producing vaccines, biologics, antibiotics, and other critical injectable products.
