

Ampoule Filler for Biological Indicator is a specialized machine designed for filling and sealing glass ampoules used in biopharmaceutical, biotech, and sterilization validation applications. These machines ensure precise filling of biological indicators, typically containing microorganisms like Geobacillus stearothermophilus spores, into ampoules for monitoring sterilization processes, such as steam or hydrogen peroxide sterilization.
Empty washed & sterilized ampoules loaded onto infeed hopper of the machine. From where it has been received to receiving rack through starwheel/cassette. The moving rack which moves horizontally collect one/two/four/eight ampoules from the receiving rack and transfer the ampoule to the machine in left to right in an inclined position. During ampoule movement Pre-gassing, Filling, Post Gassing, Pre-heating & Sealing Station perform filling and sealing operations. At the end duly filled and sealed ampoules get collected automatically onto collection tray of the machine. An Ampoule Filler for Biological Indicator is a specialized cGMP-compliant monoblock machine designed for precise filling and hermetic flame sealing of glass ampoules containing spore suspensions (such as Geobacillus stearothermophilus in nutrient broth). These ampoules are used as Biological Indicators (BIs) to validate sterilization processes like steam, ethylene oxide, and hydrogen peroxide. The machine supports both standard and onion-skin ampoules (thin-walled, crushable tubes) with gentle handling to ensure sterility and product integrity.
The machine handles ampoule sizes from 1 ml to 30 ml (model dependent), including small-volume and mini spore ampoules as well as thin-walled onion-skin ampoules. Available models: AFS-1, AFS-2, AFS-4, and AFS-8 heads.
Filling accuracy is ±1% with volumetric piston pumps, making it ideal for consistent spore population in every BI ampoule.
Nitrogen flushing (before filling, during filling, and after filling) removes oxygen and creates an inert environment inside the ampoule. This prevents oxidation of the sensitive spore suspension, avoids contamination, and enhances the stability and shelf life of the Biological Indicator. Neck-centering during flushing ensures effective purging, which is critical for maintaining the certified resistance characteristics of the spores.
