Biological Indicator Ampoule Filling Machine

Description

Biological Indicator Ampoule Filling Machine is a specialized piece of equipment designed for the precise filling and sealing of glass ampoules used in biopharmaceutical, biotech, and medical industries. These machines are critical for producing biological indicators (BIs), which are used to validate and monitor sterilization processes, such as steam or formaldehyde sterilization, by containing viable microorganisms like Geobacillus stearothermophilus spores. Below is a detailed description based on available information:

  • Sterilization: Empty ampoules are washed and sterilized, often via an infeed conveyor from a sterilizing tunnel.

  • Feeding: Ampoules are loaded into a hopper and fed onto a conveyor or slant travel system.

  • Nitrogen Purging: Pre-nitrogen flushing removes atmospheric air.

  • Filling: Precise volumes of liquid (e.g., nutrient broth with spores) are dispensed using filling needles.

  • Post-Nitrogen Flushing: Additional nitrogen flushing ensures sterility.

  • Sealing: Ampoules are pre-warmed, rotated, and sealed using gas flames or other methods.

  • Collection: Sealed ampoules are collected in a dead tray or outfeed conveyor for inspection or further processing.

Working Principle of Biological Indicator Ampoule Filling Machine

Empty washed & sterilized ampoules loaded onto infeed hopper of the machine. From where it has been received to receiving rack through starwheel/cassette. The moving rack which moves horizontally collect one/two/four/eight ampoules from the receiving rack and transfer the ampoule to the machine in left to right in an inclined position. During ampoule movement Pre-gassing, Filling, Post Gassing, Pre-heating & Sealing Station perform filling and sealing operations. At the end duly filled and sealed ampoules get collected automatically onto collection tray of the machine. Types of Ampoule Filling Machine

Applications of Ampoule Filling Machine

Benefits of Biological Indicator Ampoule Filling Machine

High Precision: Ensures accurate fill volumes and consistent sealing, critical for biological indicator reliability.

Contamination Prevention: Nitrogen flushing and aseptic design minimize contamination risks.

Efficiency: Automation and high output (up to 36,000 ampoules/hour) meet large-scale production demands.

Versatility: Handles various ampoule sizes and types, with minimal changeover time.

Durability: Stainless steel construction and cooling systems enhance longevity.

Key Features of Biological Indicator Ampoule Filling Machine

  • Purpose and Application:
    • The machine is used to fill and seal glass ampoules with biological indicators, typically consisting of bacterial spores suspended in a nutrient broth (e.g., Soybean Casein Digest Broth) with a pH indicator like bromocresol purple for detecting spore growth. These ampoules are employed to verify the effectiveness of sterilization processes in industries such as pharmaceuticals, biotechnology, and cosmetics.

  • Design and Construction:
    • Monobloc Design: Combines filling and sealing operations on a single platform, enhancing efficiency and reducing contamination risks.
    • Materials: Constructed with high-quality stainless steel (typically 316L for contact parts and 304 for non-contact parts) to meet cGMP (current Good Manufacturing Practice) standards, ensuring durability and compliance with sterile production requirements.
    • Compact and Ergonomic: Designed for easy maintenance, accessibility, and cleanability, with some models featuring tool-less format changes for flexibility.

  • Filling and Sealing Process:
    • Filling Mechanism: Operates using volumetric piston pumps, peristaltic pumps, or servo-driven systems to achieve precise filling volumes (0.5ml to 25ml in a single dose). The machine can handle ampoules ranging from 1ml to 30ml.
    • Nitrogen Flushing: Equipped with pre- and post-nitrogen flushing units to purge atmospheric air and prevent contamination, ensuring the sterility of the contents.
    • Sealing: Utilizes combustible gases (oxygen and LPG/propane) or thermal/laser methods for hermetic sealing. The sealing process includes pre-warming stations to heat the glass for perfect sealing without compromising the ampoule’s integrity. A rotation mechanism ensures uniform sealing by centering the ampoule neck during warming and sealing operations.
    • Ampoule Handling: Ampoules are fed via a hopper, transported through a wire mesh conveyor or slant travel system, and advanced in batches (1, 2, 4, 6, or 8 ampoules per stroke) for filling and sealing. A “no ampoule, no fill” system prevents wastage.

  • Production Capacity:
    • Capable of filling 10 to 250 ampoules per minute, depending on the model and configuration (e.g., single-head, four-head, or eight-head machines).

  • Advanced Features:
    • Automation: Fully or semi-automatic models reduce human intervention, minimizing errors and improving efficiency. Automated systems include PLC (Programmable Logic Controller) and HMI (Human-Machine Interface) for real-time monitoring, error detection, and parameter adjustments.
    • Sterility and Cleanliness: Some machines integrate with laminar airflow (LAF) hoods, RABS (Restricted Access Barrier Systems), or isolators to maintain aseptic conditions. Inbuilt laminar airflow or CIP/SIP (Clean-in-Place/Sterilize-in-Place) systems ensure sterile class 100 filling.
    • Pre-Heating and Cooling: Includes pre-warming stations for thick glass ampoules and cooling systems to prevent overheating, extending machine lifespan.
    • Waste Management: Features dead trays for upright collection of sealed ampoules and waste tip collectors to manage glass residues.
    • Customization: Supports filling and sealing of both open-mouth (Type B and C) and closed (Type D) ampoules, with flexibility for various neck dimensions, thicknesses, and formats.

  • Specialized Models:
    • Some machines are custom-designed for one-stop assembly and testing of biological indicator products, automating tasks like feeding and assembling ampoule caps, strips, and absorbents. These improve product quality, flexibility, and efficiency.
    • Models offer gentle handling, precise filling with multiple pump types (valve piston, peristaltic, or time-pressure), and in-process control (IPC) for weight and stopper checks.

  • Compliance and Safety:
    • Adheres to international standards like ISO, USP, and FDA guidelines for biological indicator production and pharmaceutical packaging.
