Automatic Dry Powder Filling Rubber Stoppering Machine is a specialized pharmaceutical packaging system designed for the precise filling of sterile dry powders into glass vials and immediate sealing with rubber stoppers (bungs). It combines powder dosing via a vacuum-pressure or servo-driven powder wheel with an integrated rubber stoppering unit, ensuring aseptic conditions under laminar flow or isolator technology. This monobloc (single-unit) design minimizes space in cleanrooms, reduces contamination risks, and complies with cGMP, 21 CFR, and FDA standards. It’s ideal for micro-doses of injectable powders, handling vial sizes from 5ml to 30ml with necks of 13mm or 20mm. Unlike general powder fillers, this machine uses time-dosed air/nitrogen flushing for accuracy (±1-2% variance) and features safety interlocks like “no vial/no fill” and “no stopper/machine stop” to prevent errors. It’s commonly used in high-volume sterile production lines.

The machine operates in an intermittent or continuous motion cycle, integrating filling and stoppering in a compact footprint:

 

  • Vial Infeed: Sterilized vials enter via an infeed turntable and Delrin conveyor belt, guided by separators to maintain upright orientation.

 

  • Powder Filling: Vials index to the powder wheel. Powder from the SS316L hopper is dosed using vacuum to draw it into ports, followed by timed bursts of sterilized compressed air or nitrogen to dispense exact amounts into vials. Pre- and post-gassing options flush oxygen for stability.

 

  • Vial Separation: Filled vials are separated by grooves on the conveyor and moved to the stoppering station.

 

  • Rubber Stoppering: Sterilized, siliconized rubber stoppers are oriented in a vibratory bowl, stacked in a chute, and picked/placed via vacuum or roller systems. Vials pass under pressing rollers for secure full or half-bunging.

 

  • Outfeed: Completed vials exit via an outfeed turntable, ready for capping or labeling. AC frequency drives synchronize speeds, with PLC/HMI for servo models enabling multi-dose adjustments.

 

Maintenance involves tool-free disassembly of SS316L contact parts for autoclaving. Dust extraction and HEPA filtration ensure sterility.

Working Principle of Automatic Dry Powder Filling Machine

The sterilized powder is stored in powder hopper is agitated by pair of mechanical agitators for maintaining consistency and uniform bulk density. The powder wheel having eight ports rotates at the pre determined speed below the powder hopper with practically no clearance. Powder wheel consist of Piston in each port and behind the powder wheel vacuum plate is provided there is no clearance between powder wheel and vacuum plate due to back spring pressure. Precise volume of powder is sucked in to the port of powder wheel during vacuum according to the piston length different fill size can be achieved. The excess powder is doctored off by a doctor blade. Doctor blades can be adjusted from outside also without removing powder hopper. When powder wheel indexes further and remain in the port due to the vacuum till it reaches just vertically above the container. The time dose of Compressed air (Nitrogen Gas), sterilized low pressure air sequentially flushes out powder from the port of powder wheel in to the container one by one. The filled containers are immediately separated on the conveyor by vial separator and moves further for stoppering operation. (Different grooves separator can be used which depends on the container diameter or multiple dosing systems). The sterilized, siliconised, rubber stoppers stored in the vibrator bowl moves to vibratory bowl and stacked vertically in the rubber stopper chute. The container is hold firmly between pair or timing belt to pickup rubber stopper from exit end of the chute. Further the container is passed between two pressing roller for tight fit fixing of rubber stopper.
