Sterile Preparation Vessel

Description

Sterile Preparation Vessel (also known as a Sterile Manufacturing Vessel or Aseptic Mixing Vessel) is specialized equipment used in the pharmaceutical, biotech, and healthcare industries for the safe, contamination-free preparation, mixing, and storage of sterile liquids. These vessels are critical for producing injectable drugs, eye drops, oral liquids, inhalers, and other sensitive formulations that require aseptic conditions to prevent microbial contamination. They comply with strict standards like cGMP (current Good Manufacturing Practice), USFDA, WHO, and ASME BPE guidelines to ensure product integrity and patient safety.

Working Principle of Sterile Preparation Vessel

Jacketed Mixing Vessel (double jacketed vessel) including jacketed steam boiler is uses for mixing liquid and slightly solid materials. Jacketed Mixer assembly equipped with series of paddles types blades, pitched or flat blade of appropriate shape. In Mixer two consecutive blades move in opposite direction which creates laminar thinning of the material and facilitates uniform mixing. Paddle type blades mounted on scrapping arms and welded to hollow shaft have independent drive. Bottom support to the shaft called piyot provided for firmness of shaft during rotations. Paddles mounted on internal shaft creates relative movement of the material. For discharge of material different types of valves can be offered as per user requirements.

Key Features of Sterile Preparation Vessel

Sterile preparation vessels are engineered for hygiene, durability, and precision. Common attributes include:

  • Materials: Typically constructed from high-grade stainless steel (e.g., SS 316L) for corrosion resistance and smooth surfaces (electropolished to Ra < 0.25 µm for easy cleaning).

  • Capacity Range: From small-scale (1–10 liters for R&D or high-value products like vaccines) to large industrial sizes (up to 20,000 liters).

  • Jacketed Design: Double or triple jackets for heating/cooling (via steam, hot water, or chilled fluids) and temperature control (e.g., 40–60°C to protect heat-sensitive ingredients).

  • Agitation Systems: Magnetic stirrers, paddle blades, or variable-speed agitators for uniform mixing of viscous or dense fluids without introducing contaminants.

  • Sterilization Capabilities:
    • SIP (Sterilization-in-Place) using pure steam.
    • CIP (Clean-in-Place) for automated washing.
    • Nitrogen blanketing or sparging to displace oxygen and inhibit microbial growth.

  • Filtration Integration: Inline sterile filters (0.22–0.45 micron) to remove particles and microorganisms during transfer.

  • Controls and Monitoring: PLC-based panels for pH, temperature, pressure, and dissolved oxygen (DO) monitoring; often placed in Grade B cleanrooms under positive nitrogen pressure.

  • Accessories: Flanged tops with silicone gaskets, harvesting ports, sampling valves, and variable frequency drives for stirrer speed.

These vessels are fully enclosed with domed tops and bottoms to minimize dead zones and facilitate drainage, reducing the risk of residue buildup.

FAQ for Sterile Preparation Vessel

Key features include:

  • ASME code design with SS 316/316L contact parts and electropolished surfaces.
  • Complete aseptic design with zero dead-legs, CIP/SIP compatibility, and magnetic mixers.
  • Jacketed construction for precise heating/cooling.
  • Load cell volume monitoring, pH/conductivity sensors (optional), and flush bottom diaphragm valves.
  • Top-mounted pitch-type stirrer with VFD for adjustable speed.

The double-jacketed vessel uses a top-mounted pitch-type stirrer (up to 150 RPM via VFD) for uniform mixing of liquids and semi-solids. Heating/cooling is achieved through the jacket (steam, hot water, or chilled fluids). The system supports CIP/SIP, nitrogen blanketing, and sterile filtration while maintaining an enclosed, hygienic environment to ensure product sterility.

Sterile Preparation Vessels are available in capacities ranging from 5 liters to 20,000 liters. Standard specifications include 0.5 HP to 75 HP power, 440V 3-phase supply, helical gearbox, and SS 304/316/316L stirrer material. Designs are fully customizable for specific process needs.

They are widely used in the Pharmaceutical, Biopharmaceutical, Biotech, Cosmetic, Chemical, Ayurvedic, Herbal, and Nutraceutical industries. Major benefits include superior sterility & contamination control, excellent temperature regulation, easy CIP/SIP cleaning, full regulatory compliance (cGMP, USFDA, WHO, ASME BPE), and high product integrity for sensitive formulations.
